Uncouplers are chemicals that disconnect (uncouple) electron transport from ATP synthesis.

Normally:

ETC → pumps H⁺ → forms proton gradient → ATP synthase makes ATP

With uncouplers:

– They destroy the proton gradient by allowing H⁺ ions to leak back into the matrix without passing through ATP synthase.

– ETC runs faster, consuming more oxygen.

– ATP production drops.

– Energy is released as heat (non shivering thermogenesis)

• Thyroxine

• Thermogenin (Uncoupling protein 1; UCP-1): 

found in brown adipose tissue

• Long chain fatty acid

• 2, 4 – DNP (Di nitro phenol)

• Di nitro cresol

• FCCP (Fluoro carbonyl cyanide phenyl hydrazine)

• Asprin (in high dose)
